Volume of distribution gives information about the concentration of drug present in plasma after the administration of dose. as the drug binds to the tissue components the volume of distribution can be known. If Vd is large then it means that drug is extensively binded. Ref:http://www.merckmanuals.com/professional/sec20/ch303/ch303d.html

Volume of distribution correlates the amount of the drug present in the body and the concentration of the drug present in the systemic circulation. The clearance of the drug from the body is influenced by this parameter. The clearance is the product of elimination rate constant and the volume of distribution. The dose requirement and the dosing frequency is dictated by this term.
